盛唐时期，年方二十的狄仁杰上京参加明经考试，武则天亲自监考。一名伪装成考生的刺客忽然杀出，考场大乱，狄仁杰追捕神秘刺客，意外发现刺客是女扮男装，刺客趁乱逃走。武则天命狄仁杰继续追查刺客的来历，狄仁杰发现对方的暗杀是经过详细周密部署的。年轻的狄仁杰决心不追查到底不罢休。 狄仁杰才华出众，又护主有功，很快当上了汴州判佐。在汴州，狄仁杰遇上了初恋情人楚芸。楚芸比狄仁杰大了五岁，楚芸就像姐姐一样照顾着狄仁杰。后来楚芸嫁给了左司郎中王本立的二公子王彦，新婚之夜王彦意外身亡。此时再遇狄仁杰，两人已然心动，但经历大变故的楚芸已理智地知道她和狄仁杰没有未来，而狄仁杰却发现王彦之死并不是一场意外，惨剧一幕幕发生，一连王家的几人皆死于非命，命案现场凄美得诡异，而狄仁杰与楚芸暗涌的情感为案情更添神秘。
